Mối liên quan giữa vermiform và bệnh phân trắng

Vermiform là một “vật thể” lạ được tìm thấy nhiều trong gan tụy tôm bị phân trắng.

Bề ngoài vermiform giống “một quả lựu đạn”, nằm gần như vừa khít trong ống gan tụy.

Bài viết này mô tả thể Vermiform. Đây là một “vật thể” lạ được tìm thấy nhiều trong gan tụy tôm bị phân trắng. Bề ngoài vermiform nhỏ, thấy được qua kính hiển vi bao gồm nhiều màng mỏng độc lập kết hợp lại với nhau thành một thể liền mạch. Vậy đây có phải là một mầm bệnh thực thụ trên tôm như virus, vi khuẩn, nấm hay ký sinh trùng không?

Hàng trăm mẫu tôm đã được nghiên cứu kéo dài từ 2009 đến 2012 để cố gắng xác định nguyên nhân gây ra hội chứng hoại tử gan tụy cấp tính (AHPND), đây cũng là một đại dịch nghiêm trọng gây thiệt hại nặng nề cho ngành nuôi tôm toàn Châu Á không kém gì phân trắng. Hoại tử gan tụy cấp tính chủ yếu bùng phát trên tôm sú và tôm thẻ chân trắng, lan sang Việt Nam năm 2010, khi đó vẫn chưa biết được nguyên nhân chính xác của bệnh là gì. Sang năm 2012, hội chứng này lại xuất hiện trên một tạp chí thủy sản uy tín với tên gọi “hội chứng tôm chết sớm (EMS)”. Cuối cùng đến 2013 thì tác nhân gây bệnh mới được chỉ đích danh là vi khuẩn vibrio parahaemolyticus, với một đặc điểm duy nhất là dịch bệnh này lây lan ồ ạt và gan tụy tôm nhiễm vi khuẩn bị bong tróc ở các tế bào phần giữa lớp biểu mô.

Trọng tâm chính của hội chứng AHPND là gan tụy với hàng trăm đoạn mô đã được kiểm tra. Trong số những điểm bất thường, càng kiểm tra thì các chuyên gia càng phát hiện có nhiều thể vermiform, bề ngoài giống “một quả lựu đạn” nằm bên trong ống gan tụy trên cả tôm sú và tôm thẻ chân trắng. Khi đã đạt đủ số lượng cần thiết thì sẽ làm xuất hiện các chuỗi phân trắng trong ao tôm từ tháng thứ 2 trở đi. Và các thể vermiform này bắt đầu bị nghi ngờ là căn nguyên chính của hội chứng phân trắng (WFS). Khi phân trắng nổi đầy trên bề mặt ao, nếu tôm không chết thì lại trở nên còi cọc, chậm lớn. Với con số ngất ngưởng thế này thì mức độ thiệt hại còn nặng hơn rất nhiều so với bệnh đốm trắng trước đây.

Ở đây, vermiform được mô tả chi tiết về cấu tạo và hình dạng. Chúng không độc lập mà tập hợp thành tập đoàn, do các vi nhung mao biến đổi, bong tróc ra từ các tế bào biểu mô ống gan tụy tôm mà thành. Vermiform còn có thể tích tụ trong cả đường ruột trước khi được thải ra ngoài theo phân. Nhiều mẫu tôm còn không có vẻ gì là đã nhiễm bệnh nhưng vẫn phát hiện được nhiều thể vermiform trong gan tụy.

Khi đã mắc phân trắng, đường ruột tôm sẽ chứa đầy phân từ trắng đến vàng và bị phồng lên. Dưới kính hiển vi quan sát được nhiều thể vermiform có hình dạng bên ngoài giống như ký sinh trùng gregarine. Đến 96% tôm nhiễm phân trắng được phát hiện trong gan tụy chứa rất nhiều thể vermiform. Ở các ao nhiễm bệnh, tỷ lệ sống giảm 20-30% so với các ao bình thường. Mức độ tiêu thụ thức ăn và tăng trưởng cũng giảm đáng kể, mức tăng cân bình quân mỗi ngày cho cả vụ ít hơn 0,1g/ngày so với con số 0,2g/ngày ở các ao bình thường. Hệ số chuyển đổi thức ăn (FCR) cũng dao động từ 1,7 đến 2,5 so với 1,5 hoặc ít hơn ở những ao không bệnh.

Các mẫu kiểm tra ở bất kì giai đoạn nào kể cả tôm bố mẹ đều có sự hiện diện của vermiform, với kích thước gần như khớp với các ống gan tụy mà chúng tồn tại. Vermiform không hề có tế bào và cũng không có thành phần nào có đặc điểm của tế bào, mà chỉ chứa những “tấm màng” mỏng là những mảnh rải rác xen kẽ các thành phần bị bong tróc, kết hợp và ngưng tụ trong 1 lớp bao bọc bên ngoài. Sau đó nhiều thể vermiform cùng tích tụ tại trung tâm các ống gan tụy.

Trước khi có nghiên cứu này thì vermiform được mô tả là giống với gregarine về cấu tạo và hình dạng. Tuy nhiên, chưa có tài liệu nào kết luận gregarine giống với kết quả mô tả vermiform ở hiện tại. Cụ thể vermiform không thể vận động và thiếu các bào quan để cấu thành cơ thể. Chúng chứa nhiều lớp mỏng bong tróc từ gan tụy, giống các lớp giấy bóng kính, xuất phát từ vi nhung mao của tế bào biểu mô ống. Chứ không hề giống với với cấu tạo của một ký sinh trùng. Quá trình bong tróc gan tụy chỉ ra rằng sự hình thành vermiform là do bệnh lý.

Khi xuất hiện quá nhiều vermiform có thể dẫn tới việc hình thành các chuỗi phân trắng ở tôm. Điều quan trọng là tỷ lệ gia tăng của chúng lại tỷ lệ thuận với sự xuất hiện của AHPND hay các bệnh khác như nhiễm khuẩn vibrio, vi bào tử trùng Enterocytozoon hepatopenaei. Kết quả làm nguyên nhân gây phân trắng trên tôm ở Việt Nam từng được cho là do Enterocytozoon sp, nhưng điều này đã được chứng minh là vô lý.

Các biện pháp an toàn sinh học ở các hệ thống nuôi chưa được chặt chẽ, chưa đủ sức chống lại sự xâm nhập của mầm bệnh bên trong lẫn bên ngoài. Do đó, có hai khả năng, một là vermiform tạo ra bởi các tác nhân từ bên ngoài, hai là do các mầm bệnh có sẵn trong cơ thể tôm đã gây ra cùng lúc với một số bệnh tôm đã mắc trước đó. Ví dụ như vi khuẩn gây AHPND có thể tiết ra độc tố làm bong tróc tế bào biểu mô ống gan tụy và tạo thành vermiform.

Như vậy có thể kết luận rằng những trường hợp bị phân trắng nghiêm trọng là do sự hình thành quá nhiều vermiform trong cơ thể tôm. Bệnh phân trắng dĩ nhiên sẽ tác động tiêu cực đến sự tăng trưởng và tỷ lệ sống của tôm. Tuy nhiên nguyên nhân hình thành vermiform thì vẫn còn là một ẩn số. Hy vọng rằng sau khi hiểu được bản chất và phương thức hình thành của “vật thể” này sẽ có hướng để xác định nguyên nhân chính xác tạo ra chúng.
